Macromolecular Concentrations in Articular Cartilage by Fourier Transform Infrared Imaging

چکیده

INTRODUCTION: Fourier transform infrared imaging (FTIRI) is a spectroscopic imaging tool that can be used to study the compositions and structures of biological tissues with fine spatial and spectral resolutions. Many FTIRI studies of articular cartilage use the peak intensities/areas in the infrared absorption images to represent the molecular contents, which can contain large error due to the considerable overlaps of the characteristic bands of multiple molecular components in biological tissues.

Chemical visualization of individual chondrocytes in articular cartilage by attenuated-total-reflection Fourier Transform Infrared Microimaging

Fourier transform infrared imaging (FTIRI) and the attenuated total reflection Fourier transform infrared microimaging (ATR-FTIRM) were used to study the chemical and structural distributions of cellular components surrounding individual chondrocytes in canine humeral cartilage, at 6.25µm pixel resolution in FTIRI and 1.56µm pixel resolution in ATR-FTIRM. The chemical and structural distributio...

Fourier-transform infrared anisotropy in cross and parallel sections of tendon and articular cartilage

BACKGROUND Fourier Transform Infrared Imaging (FTIRI) is used to investigate the amide anisotropies at different surfaces of a three-dimensional cartilage or tendon block. With the change in the polarization state of the incident infrared light, the resulting anisotropic behavior of the tissue structure is described here. METHODS Thin sections (6 mum thick) were obtained from three different ...

Fourier Transform Infrared (FTIR) Microspectroscopy of Immature, Mature and Degenerated Articular Cartilage

Osteoarthritis is a common joint disease characterized by the progressive degeneration of the articular cartilage. In this chapter, the application of Fourier transform infrared (FTIR) microspectroscopy for the determination of spatial composition of main structural components of articular cartilage, i.e. collagen and proteoglycans, is presented. The previous results from FTIR microspectroscopi...

Fourier Transform Infrared Imaging of focal lesions in human osteoarthritic cartilage.

Fourier Transform Infrared Imaging (FTIRI) is a new method for quantitatively assessing the spatial-chemical composition of complex materials. This technique has been applied to examine the feasibility of measuring changes in the composition and distribution of collagen and proteoglycan macromolecules in human osteoarthritic cartilage. Human cartilage was acquired post-operatively from total jo...

Specificity of Fourier Transform Infrared (FTIR) Microspectroscopy to Estimate Depth-Wise Proteoglycan Content in Normal and Osteoarthritic Human Articular Cartilage

BACKGROUND Fourier transform infrared (FTIR) microspectroscopy is a promising method for estimating the depth-wise composition of articular cartilage. The aim was to compare the specificity of two earlier introduced, presumably proteoglycan (PG)-specific FTIR parameters (i.e., absorption in the carbohydrate region with and without normalization with Amide I absorption) to estimate the reference...
